i want to remove Tween 8o from a Human tPA protein. I know i can do it by dialysis . How can i actually do that and most imp which buffer i should use??????
Hi Mohammad
Tween 80 (http://en.wikipedia.org/wiki/Polysorbate_80) is nonionic and can be removed from charged protein and peptide samples by ion exchange chromatography. The choice of resin and buffer depends on protein amount and subsequent analysis. For mass spectrometric analysis at Alphalyse we typically use SCX material, for example SCX ZipTips from Millipore, with loading, washing, and elution buffers as described by Millipore.
